Natural products down plants against pseudomonas aeruginosa and staphylococcus aureus: antibiophilm activities and molecular mechanisms

Natural products down plants against pseudomonas aeruginosa and staphylococcus aureus: antibiophilm activities and molecular mechanisms

Bacteria are social organisms that are able to build complex structures, such as biofilms, namely a very organized surface microorganism community, wrapped in its own extracellular matrix. Biofilms are usually related to many health problems because their formation increases resistance to antibiotics and antimicrobial agents, such as in the case of pseudomonas aeruginosa and staphylococcus aureus, two human pathogens that cause the main concern. P. Aeruginosa is responsible for severe nosocomial infections, the most often includes ventilator related pneumonia, while S. aureus causes several problems, such as skin infections, septic arthritis, and endocarditis, for only a few.

Literary data shows that natural products from plants, bacteria, fungi, and marine organisms have been shown to be effective as an anti-biofilm agent, inhibiting the formation of polymer matrices, suppressing cell adhesion and attachment, and reducing the production of virulence factors, thus blocking the quorum sensing network , Here, we focus on plant derivative chemicals, and provide literature reviews that are updated on the nature of anti-biofilms, flavonoids, alkaloids, and phenolic compounds. In addition, every time information is available, we also report the mechanism of action.

Bioactive Chrysin and Luteolin flavonoid protection measures on the formation of glyoxal of the final product of advanced gliking and human serum albumin aggregation: in vitro and molecular docking analysis

Modification of post-translational proteins by nonenzymatic (neg) and age accumulation are two underlying factors associated with long-term pathogenesis in diabetes. Glyoxal (GO) is an intermediary reactive that has the ability to modify proteins and produce age at a faster level. Human Serum Albumin (HSA) becomes the most abundant serum protein has a higher opportunity to be modified by the neg. The main objective of this study is to investigate the potential of Chrysin and Luteolin as an antigry and antifibrillation agent in mediated glication and the formation of HSA mediated. The formation of age is confirmed from spectral measurement of absorption and fluorescence.

Both flavonoids are able to satisfy the intensity of fluorescence in age in vitro indicating antiglica properties of molecules. Fibril formation in the modified HSA is confirmed by Thioflavin T (ENT) and flavonoids fluorescence tests found to provide antifibrillation properties in vitro. The results of the docking suggest that both flavonoids interact with various amino acid residues of iia subdomain, including licens are vulnerable to glication and arginine through non-covalent strength and further stabilize the HSA structure, which further explains their action mechanism as an antiglicate agent and antifibrillation.

Comprehensive quality evaluation of Fuzi and its processed products through UPLC-QTOF / MS integration combined MS / MS based spectral molecular networks with multivariate statistical analysis and HPLC-MS / MS.

Aconiti Lateralis Radix Praeparata (Chinese name is Fuzi, FZ), lateral root or son of asonitum Carmichaelii debx. (Ranunculaceae), is a controversial traditional Chinese medicine (TCM) distributed universally and applied in many countries, such as China, Japan, Korea and India. FZ can be used to treat various diseases, including rheumatic fever, rheumatism, painful joints, syncope, collapse, bronchial asthma, some endocrinal disorders, etc. However, the quality control and assessment of FZ challenges due to clear and high toxicological risks, and only processed products are permitted clinically used in accordance with relative safety regulations. As a result, it is necessary to analyze all chemical compositions and dynamic FZ changes before and after processing. Overcoming chemical changes from raw and processed products is a way to reduce toxicity.

Natural products down plants against pseudomonas aeruginosa and staphylococcus aureus: antibiophilm activities and molecular mechanisms
The purpose of this study: In this article, the entire Chemical Composition of FZ was analyzed, the difference between FZ RAW and processed was evaluated, and the possibility of factors that influenced the decrease in processed FZ toxicity described from the perspective of its chemical composition using qualitative and quantitative analysis methods.

Materials and Methods: New strategies of many data collection and processing based on ultra-performance liquid chromatography coupled with the Mass Spectrometry method for quadrupole time (UPLC-QTOF / MS) in positive ion mode, along with the natural product of the global social molecular network (GNP) and analysis Multivariate Statistics, was established to systematically identify the chemical constituents of FZ and investigate comprehensively chemical markers that can be used to distinguish FZ processed with vinegar and honey from raw products. Combined with the results of qualitative analysis, 12 components, including 8 chemical markers compounds and 4 toxicity components, are analyzed quantitatively by using high-performance liquid chromatography equipped with triple-quadrupole mass spectrometry (HPLC-MS).

Results: Using the molecular network analysis method (MN), a total of 145 compounds identified, where 13 was identified using reference compounds. Seventy-seven chemical markers are also detected between raw and processed FZ. The results of identification of the markers of the chemicals are also verified by the analysis of orthogonal partial partial discriminant (OPLS-DA). Quantitative results indicate that the contents of 12 important components are all decreased, especially the diagester of alkaloid (ddas), after processing.
